The in-vitro micronucleus assay is a globally used technique for the quantification of DNA damage induced by chemical, radiation or some nano-materials and is required for regulatory safety testing in addition to inter-individual monitoring of environmental, lifestyle and occupational factors. The assay is the gold-standard test of genotoxicity in the development of all chemicals/pharmaceuticals. When the nucleus divides during mitosis, chromosome fragments that fail to be incorporated into the daughter nuclei appear as micronuclei within the cell (Fig.1E). Previously we showed that Imaging flow cytometry is an effective measurement tool for the micronucleus assay given the high-throughput single-cell nature of the data. We also developed a deep learning algorithm to remove the user-subjective manual scoring of individual cell images.
There are specific rules for scoring micronuclei events in terms of the nuclear size, intensity of the DNA stain and we would like to train neural networks which incorporate the scoring rules which are accepted by the community. This will allow us to develop assays which will be adopted by the community. |
